Can I use six regular 12 volt batteries in place of the six volt batteries on my golf cart?

Not unless you change it to a 12 volt system , you will blow it up , the electric motor is set up for 6 volt power, and will be run over itself with this much battery power,

Question details are not sufficient for perfect answer. Doubling the operating voltage will force the double amount of current thereby burning the motor.
